The same thing happened to me a few times and then found this Rockstar support page

I fast tracked to Armadillo, saved the game and then reloaded that save, then fast tracked back to Las Hermanas. I then just stood still for about 10 seconds to let everything load and then crouched and slowly walked into the church where Mother Superior mission marker was and all was fine.

managed to complete the mother superior blues mission but it froze on me while inside the church in Las Hermanas aswell ,so what ive done is delete all the game data & reinstall it & i also walked into the las hermanas area,as everytime i was on a horse it would freeze,but tbh i shouldnt have had to do this if the dlc was ok in the first place

The post from WezFx totally worked for me! I also used the idea from the R* link to come to the church from a different direction. From the NW seemed to be it....the undead are now dead.

I know this won't help but FYI I have completely the mission for the nun at Las Hermanas without any freezing issues.
As soon as I crossed into Mexico I was notified that Blackwater had been taken over so I crossed the river back and I transported from Plainview to Blackwater. Then I returned to Plainview, crossed the river and completed the mission for the nun. I guess that additional work saved me from the problem.

I have this sae problem, but my game crashes on the train to Mexico. Just after John has smashed through the debris on the track it freezes and I still hear sound until the cutscene is over. My whole PS3 crashes, not even the PS button works.

I obviously talked too soon. I have had at least 3 or 4 crashes in the last couple of days. Always around Las Hermanas. One of the missing persons one needs to save was actually hiding inside Las Hermanas and I had to try three times before I could manage to enter the monastery without the PS3 freezing.Even after the completion of the story there are still several things to do in Mexico so it's easy to pass by Las Hermanas and crash. I hope they come up with a correction SOON.

finished the main game- its when playing as a zombie

dont think it matters if your playing as a zombie or not m8,i`ve had problems before & after completing the game,its a shame the dlc is so buggy for some,hope rockstar are working on a patch for it

There are also other areas in the DLC that are problematic. When approaching Fort Mercer (and to a smaller extent Armadillo) the game freezes for a second and then resumes. Luckily it doesn't freeze completely.
I have a feeling that the game needs to fetch data for the location (buildings, roads, etc) from the BluRay disc and at the same time it must overlay it with additional information from the DLC i.e. from the hard disk and this is causing some kind of timing problem.
