Oh sorry, I mean't the Platinum trophy.

As the others have said, you can get the Platinum after two playthroughs... but it depends how you want to go about it, and what you want to walk the plank o' your playthroughs...
I'm a fan o' not focussing on bonus trophies while playing the hardest difficulty... I like to have everything else done before attacking Crushing, Superhuman etc... thar is no way you can get all the other trophies on one play... unless you deliberately die, so that you can ho trophies at the same spot o' the game, like stealth kills on the same pirate with his back turned to you... or deliberately die so you can get enough kills from each weapon.... that to me is no fun, and defeats the purpose o' a linear story game.... I'd rather just play the game through one more time, or at the very least, level select areas like Act 4 where I know I can farm the hangman medals, the explosion medals, the combo medals, and even the stealth blow the son of a biscuit eater down medal...
Resistance 2 I actually enjoyed more on the harder two difficulties, than the easier two... because I wasn't trying to get trophies, and was playing it with the weapons that the game designers want you to use, and when... because it took me four runs, I was starting to tire o' the game on my Superhuman run though..
So... I'm a fan o' three playthroughs in any story based game, if I'm enjoying it.... a fourth, well... can sometimes be too much.... I'd have no desire to play Dead Space a 4th time... Uncharted is the exception o' a game where I wanted to play it after three runs.
RE5 was different because it was a DMC style chapter select game.... with the exception o' the first co-op playthrough on the easiest setting with Smelck, I didn't play the game linearly through all 4 difficulties... I played Normal for a week while I waited for Smelck to come back online to finish Easy... then I'd level select Easy by myself to try and S rank levels, while playing some Normal, and playing some Veteran... Then I'd level select a lot to farm myself up to finishing off all the time consuming Silver trophies... before finally in one last swoop tackling Professional in one linear run... some o' it with Smelck, but most o' Pro I did by myself.. Even then though after 50 hours and deciding I needed a long break from the game, I played it again with him this weekend to help him advance his Veteran levels..
In summary... I like to try and play once as naturally as I can... just letting the trophies come to me, while swallowing in the story, with no maps or jumping on the web for advice... (perfect example would be Dead Space.. why would you do a cutter only run on your first play just for the trophy?.. you are denying yourself the fun o' unlocking a large range o' weapons when your mind is freshest to the game)..... then I'll attempt a second run, and try and pickup all the rest o' the trophies... then hopefully a third run that focusses on just beating the hardest difficulty..

Pretty much how i go about it. I like to have no distractions on the hardest difficulty, so i can just focus on playing.

Whoops. That's what I'm doing now. I dunno why I've decided to do that, but I'm finding it really fun. The Cutter is just an awesome weapon. I'm really loving the game though, so when I play through on the harder difficulties, I'll definately be taking advantage o' the four weapon slots.
